PI3K 信号通路控制调节性T细胞种群

  Foxp3阳性的调节性T细胞(Treg, regulatory T cells)在维持机体免疫系统功能平衡重具有至关重要的作用。传统T细胞(Tconv, conventional T cells)和调节性T细胞之间的一项显着差别就是PI3K信号通路的活性。静息态的T细胞中PI3K信号通路由负调控因子PTEN抑制,不会被激活,而当Tconv细胞被激活时PTEN的活性会被下调,但是Treg细胞不会。由此研究者们发现控制Treg细胞中的PI3K信号通路对其种群的的体内平衡和稳定性是不可或缺的。
  研究者们使用Treg细胞特异性Pten基因敲除的小鼠建立了一种自身免疫性---淋巴增生的疾病模型,而该患病小鼠表现出过量的1型辅助性T细胞(TH1, T helper 1 cell)反应以及B细胞活化。Treg细胞中缺乏对PI3K信号通路的压制,从而导致白介素-2受体 亚基CD25的低表达,以及Foxp3阳性CD25阴性的细胞群体聚集,最终这些细胞中的Foxp3表达会完全丧失。
  总的来说,本文的数据表明通过PTEN来遏制PI3K信号通路在Treg细胞中的活化对于维持Treg细胞种群的体内平衡、免疫功能和稳定性都是非常关键的。

Here we found that control of PI(3)K in Treg cells was essential for lineage homeostasis and stability. Mice lacking Pten in Treg cells developed an autoimmune-lymphoproliferative disease characterized by excessive T helper type 1 (TH1) responses and B cell activation. Diminished control of PI(3)K activity in Treg cells led to reduced expression of the interleukin-2 (IL-2) receptor α subunit CD25, accumulation of Foxp3+CD25? cells and, ultimately, loss of expression of the transcription factor Foxp3 in these cells. Collectively, our data demonstrate that control of PI(3)K signaling by PTEN in Treg cells is critical for maintaining their homeostasis, function and stability.
